Abstract

Nucleotide sequence of hypervariable fragment of VP2 of infectious bursal disease virus (IBDV) isolated in Bali and Indonesia were compared to standard virulent (svIBDV’s) and very virulent (vvIBDV’s) viruses accessed in GeneBank. The comparation were done using the multiple sequence alignment program of Mega 3.1. The sequence were aligned by the ClustalW Method (Mega 3.1), that is grouping the sequences into clusters by examining the distance between all pairs. The sequences were aligned pair wise, then as groups. Phylogenetic tree were constructed using Neighbor-Joining and Bootstrap-tested. The analysis showed that IBDV can be clustered into 2 major groups, i.e. America-Europe and Australia clusters. The America-Europe cluster is further divided into two sub-clusters, i.e. classic IBD and vv-IBD. Classic-IBD is represented by standard STC, Cu-1, Variant A, F52-70, and PBG98 viruses. Two IBDV’s isolated in Bali and the most of other Indonesian isolates belong to sub-cluster vv-IBD and share a common cluster with IBDV that were recently reported as very virulent strains. One exceptional isolate is Indo 13, which is groupped into classic IBD, and closely related to classical American standard virus, STC.

Abstract

The aim of the experiment was to investigate the intensity of signal secreted by embryos for their survival in the early pregnancy. Embryos at morulla and blastocyst stages generated by in vitro fertilization were frozen and then thawed for in vitro culture in tissue culture medium. The protein secreted by embryo were detected 48 hours after thawing for those frozen at morulla stage, and 24 hours and 72 hours after thawing for those frozen in the blastocyst stage. Results showed that preimplantation goat embryos secreted an average of 3,041 µg protein per ml medium for the embryo cultured in tissue culture medium (TCM) 199 + 0,1 % BSA and 1,992 µg protein per ml medium for embryo cultured in TCM without BSA. Non growing embryos secreted only a small amount protein, i.e. 0,434 µg protein per ml medium and 0,417 µg protein per ml medium respectively for embryo cultured TCM 199 with BSA and TCM without BSA. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) analysis showed protein bands with many different molecular weight of 100, 95 , 55 , 43 , 28 and 18 KD. It appeared that the growing embryos secreted more proteins as compared to those non growing embryos.

Abstract

Oxidative stress as consequence of intense physical exercises cause tissue lesions. The objective of this study was to identify the comparable antioxidant effect of water extract and syrup of Balinese purple sweet potato to the liver tissue in mice after maximal exercise, through the quantification of blood and liver malondialdehyde (MDA), and the histologic feature of liver tissue. Subjects of this study were 60 adult male Swiss mice divided into 6 groups with control group post-test only design. Blood and liver were collected after exercise protocol of exhausting swimming in ten mice without water extract, ten mice with water extract and ten mice with syrup of Balinese purple sweet potato. The MDA was quantification with TBARS method. Samples were also collected from control group of ten mice without water extract, ten mice with water extract and ten mice with syrup of Balinese purple sweet potato as sedentary mice. The results show a significant increase of MDA in the blood and liver, after swimming with and without water extract or syrup of Balinese purple sweet potato respectively (p<0,05). The increase of MDA was slightly significant in the group with water extract or syrup of Balinese purple sweet potato (p<0,05). The increase of liver cell necrosis was slightly significant in the group with water extract of Balinese purple sweet potato (p<0,05). From this finding it can be concluded that water extract or syrup of Balinese purple sweet potato has antioxidant effect in exhausting exercise in mice. Water extract of Balinese purple sweet potato has protective effect to the liver cell in exhausting exercise in mice

Abstract

The aim of this study was to determine the effect of high dose calcium carbonate in rat (Rattus norvegicus) following ovaryohysterectomy. A total of twenty female rats at 13 week-old were used in this study. Following ovaryohitectomy, the animals were randomized in four treatment groups. Group P0 were :fed with standard food only P1, P2 and P2 groups treated with standard food but supplemented calcium carbonate respectively at the dose of 75 mg per animal per day, 225 mg per animal per day , and 450 mg per animal per day. The calcium carbonate supplement were given daily in the morning for 42 days. The experimental animals were sacrificed at 21 week-old. Calcium and phosphor level in sinister kidneys were determined by spectrofotometric method. The data obtained from this study were analysed using one way analysis of variance. No significant difference was observed in calcium level among four treatment groups, with the lowest level were found in P3 group. However, the phosphor level of P1 was significantly lower than those of P2 and P3 groups. The highest phosphor level was observed in P3 group, indicating a phosphorous retension and the signs of renal failure.

Abstract

A study was carried out to isolate and characterize lactic acid bacteria (LAB) from the Sumbawa mares milk The Isolation of LAB was conducted in Man Rogosa Sharpe (MRS) agar. The isolates were characterized by standard methods, such as Gram staining, cell morphology study and fermentation activities. The ability of the isolates to inhibit some pathogenic bacteria was studied by dual culture assay. Isolates showing the widest spectrum of inhibiting pathogenic bacteria were further identified using API 50 CHL. The results showed that Sumbawa mare milk was dominated by lactobacilli and weisella/leuconostoc. As many as 26 out 36 isolates belong to homofermentative lactobacilli and another 10 isolates belong to both heterofermentative lactobacilli and weissella or leuconostoc. Twenty four isolates inhibited the growth of Escherichia coli 25922, Shigela flexneri, Salmonella typhimurium, and Staphylococcus aureus 29213. Two promising isolates with the widest spectrum of inhibiting pathogenic bacteria, Lactobacillus sp. SKG34 and Lactobacillus sp. SKG49, were identified respectively as Lactobacillus rhamnosus SKG34 and Lactobacillus ramnosus SKG49. These two isolates were specific strains of the sumbawa mare milk and are very potential to be developed as probiotic for human.

Abstract

A study was conducted to determine the pathogenicity of Escherichia coli isolated from chickens. Three isolates, Kalasan (Ec/Kls/4/02), Sleman (Ec/Sl/1/02), and Wonosari (Ec/Wno/2/02) were firstly selected and inoculated into broiler chickens at 15 days of age. Two isolates, Kalasan (Ec/Kls/4/02) and Sleman (Ec/Sl/1/02) which showed clear clinical signs and macroscopic lesions, were cultured in Brain Hearth Infussion (BHI) media for 24 hours at 37oC. Serials ten fold dilution of the purified colonies, starting from 10 1 to 10 10 was prepared and 0,5 ml bacterial sample from each dilution were inoculated intraperitoneally into 5 chickens of 15 day-old. The inoculated chickens were monitored for seven days to observe the clinical signs and microscopic lesions. The infective dose (ID)50 of each isolate was determined by Reed and Muench method. The result showed that the two isolates were pathogenic to 15 day-old broiler chickens with specific lesions of pericarditis, perihepatitis, peritonitis, and airsacculitis. Their infective dose-50 was 2 X 10 2,6 bacterial cells per ml for Kalasan isolate and 2 X 10 1,8 bacterial cells per ml for Sleman isolate. It appeared that Kalasan isolate was more pathogenic than Sleman isolate.

Abstract

Javan rhinoceros (Rhinoceros sondaicus) and Javan bull (Bos javanicus) in National Park Ujung Kulon are protected wild animals. A study to examine the presence of trematode eggs in their fecal samples was carried out. The fecal samples were collected based on the transec route in National Park Ujung Kulon. The presence of eggs in fecal samples was examined by filtration method. Trematode eggs were found in 56% of Javan rhinoceros fecal samples and 73,91% of Javan bull fecal samples.. Base on the size of the eggs it was determined that Javan bulls in National Park of Ujung Kulon were infected by Fasciola spp (17,39%) and Paramphistomum spp (56,52%). Javan rhinoceros in National Park of Ujung Kulon was infected with Fasciola spp (44%) and Schistosoma spp (12%). This study clearly showed that tramatode infection is commom among wild ruminants in Ujung Kulon National Park and attention should be taken to improve the health status of the animals especially Javanese rhinoceros and Javanese bull
